Description:
Recombinant dolphin fish (Coryphaena hippurus) growth hormone (dfGH) produced in E. coli (21.4 K) is a single, non-glycosylated, polypeptide chain containing 188 amino acids. Recombinant dolphin fish growth hormone was purified by chromatographic techniques, according to Paduel et al., Protein Expression and Purification 16, 417–423 (1999).

Physical Appearance:

Formulation:
The recombinant dolphin fish growth hormone was lyophilized from a concentrated (1mg/ml) solution with 0.02% NaHCO3.
Solubility:
It is recommended to reconstitute the lyophilized recombinant dolphin fish growth hormone in sterile 0.4% NaHCO3 adjusted, not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Stability:
Lyophilized recombinant dolphin fish growth hormone although stable at room temperature for several weeks, should be stored desiccated below -18 C. Upon reconstitution of recombinant dolphin fish growth hormone at > 0.1 dfGH mg/ml and up to 2 mg/ml and filter sterilization recombinant dolphin fish growth hormone can be stored at +4C for 2-3 weeks.
Purity:
The purity of recombinant dolphin fish growth hormone is greater than 98.0% as determined by:
(a) Analysis by reducing and non-reducing SDS-PAGE gel.
(b) Gel-filtration chromatography under non denaturing conditions.
Amino Acid Sequence:
The sequence of the first ten N-terminal amino acids of recombinant dolphin fish growth hormone was determined and was found to be Met-Gln-Ala-Ile-Thr-Glu-Ser-Gln-His-Leu.
Dimers and Aggregates:
The purified recombinant dolphin fish growth hormone consists of > 95% monomers as determined by gel-filtration chromatography.
Biological Activity:
Binding assays of the 125I-labeled PLR’s recombinant dolphin fish growth hormone to dolphin fish liver microsomal fraction resulted in high specific binding characterized by a Ka of 0.77 nM21 and a Bmax of 285 fmol/mg microsomal fraction protein. The purified dfGH was capable of stimulating proliferation of FDC-P1-B9 cells transfected with rabbit growth hormone (GH) receptor. The maximal effect of dfGH was identical to that of human GH but their respective EC50 values were 28 nM versus 0.095 nM.
Endotoxin:
Less than 0.1 ng/µg (IEU/µg) of recombinant dolphin fish growth hormone
Protein content:
Protein quantitation of recombinant dolphin fish growth hormone was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.595 as the extinction coefficient for a 0.1% (1mg/ml) solution at pH 8.0. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
Usage:
The recombinant dolphin fish growth hormone is offered by PLR for laboratory research
